What happened?
Zealand Pharma reported topline findings from ZUPREME-2, a randomized, double-blind, placebo-controlled US trial of 220 adults living with overweight or obesity and type 2 diabetes. Participants received one of three weekly petrelintide doses or placebo alongside diet and activity support. At 28 weeks, average weight loss ranged from 7.4% to 9.2% across the treatment arms, compared with 2.0% on placebo, using the efficacy estimand.
Blood-glucose measures also moved in a favourable direction. HbA1c fell by up to 0.65 percentage points with petrelintide, while it increased by 0.23 points in the placebo group. The reported placebo-adjusted difference ranged from 0.61 to 0.88 percentage points. Gastrointestinal adverse events led to discontinuation in 1.9% of petrelintide recipients and 1.7% of placebo recipients.
Petrelintide is a long-acting analogue of amylin, a hormone released alongside insulin after eating. Amylin signalling contributes to fullness and food intake regulation. These are sponsor-reported topline results, not yet a complete peer-reviewed publication, and the treatment remains investigational.
